Dosing Guide

DSIP Dosage Chart & Timing Guide.

Lying awake at 2 a.m. gets old fast. The usual DSIP dosage is 100–300 mcg before bed, but that number did not come from a trial: every human study gave DSIP by IV, at about five times that amount. Here is the chart, the units to draw, and where the numbers came from.

The short answer

250 mcg subcutaneously in the evening: 12.5 units on a U-100 syringe from a 10 mg vial mixed with 5 mL of bacteriostatic water. Start at 100 mcg (5 units), run it for two weeks, and keep a sleep log. These are clinic conventions; the trials used about 1.5 mg by IV.

DSIP Dosage Chart

Units assume a 10 mg vial in 5 mL bacteriostatic water (2,000 mcg/mL, 1 unit = 20 mcg). Different vial? Use the units table or the calculator.

Conservative start
100 mcgper injection · 5 units
FrequencyNightly, before bed
Weekly total0.7 mg/week
Cycle length1–2 weeks, then reassess

Start here if you are sensitive to anything that touches sleep, or if you take other sleep aids.

Why this dose+

DSIP dose response in animals is U-shaped: past a point, more did less. Starting low tells you whether you respond at all before you spend the rest of the vial.

MOST COMMONStandard
250 mcgper injection · 12.5 units
FrequencyNightly, before bed
Weekly total1.75 mg/week
Cycle length2 weeks, then reassess

The figure most clinics and dosing guides settle on. Judge it on your sleep log, not on how you feel the first night.

Why this dose+

No trial produced this number. It is about 17% of the 25 nmol/kg IV dose (roughly 1.5 mg for a 70 kg adult) used in the 1980s sleep trials, and it is given under the skin instead of into a vein.

Upper end
300 mcgper injection · 15 units
FrequencyNightly, or 5 nights on / 2 off
Weekly total1.5–2.1 mg/week
Cycle lengthUp to 4 weeks

The ceiling of the common range. Going higher is not a documented way to rescue a non-response.

Why this dose+

Human trials never ran longer than about 15 days, so a 4-week course is already past anything that was studied. Two doses a day also failed in the trials, so do not split or stack doses.

More is not better with DSIP
  • Animal studies found a U-shaped dose response: past a point, higher doses did less.
  • Two injections a day failed in the human trials, while one a day worked.
  • If 300 mcg does nothing after two weeks, the honest read is that DSIP may not suit you.

How many doses in a vial? A 10 mg vial gives 100 doses at 100 mcg, 40 at 250 mcg and 33 at 300 mcg. A 5 mg vial gives 33 doses at 150 mcg, not the 66 some guides quote.

Where the DSIP Dosage Came From

Most guides skip this part. Every human DSIP study gave it intravenously, at 25–35 nmol/kg, slowly over a few minutes. With a molecular weight of 848.8, 25 nmol/kg is 21.2 mcg per kg: about 1.49 mg for a 70 kg adult and 1.91 mg at 90 kg.

What the FDA said in 2026
  • FDA staff reviewed DSIP (as "emideltide") for compounding and found no human data for subcutaneous use, for either effect or absorption.
  • They counted 209 people ever dosed with it, all by IV, for 1 to 15 days.
  • They called the insomnia evidence "inconclusive and at best preliminary".
  • On July 24, 2026 the advisory committee voted 6 to 7 against allowing it to be compounded.

The positive trials all came from one Swiss group, and the two independent placebo-controlled trials were negative. That does not mean DSIP does nothing for you. It means the 100–300 mcg range is a starting point people chose, and your own sleep log is the only test that applies. For what DSIP is and how it is thought to work, see our DSIP peptide guide.

Missed a night? Skip it. DSIP clears in minutes, so a late or doubled dose does not make up for it, and two doses in one day failed in the trials.

Mixing, Injecting and Timing in 6 Steps

1
Swab both vial tops

Alcohol swab the DSIP vial and the bacteriostatic water vial and let both dry before you pierce them.

2
Add 5 mL to a 10 mg vial

Run the water slowly down the inside wall. 5 mL gives 2 mg/mL, so 250 mcg is 12.5 units: big enough to measure, and below the solubility limit that a 2 mL mix can exceed.

3
Swirl, don't shake

Roll the vial until the liquid is completely clear. If it stays cloudy or has flecks, do not inject it.

4
Dose in the evening

In the IV trials, sleep improved from the second hour after the injection, after a slightly alert first hour. Many people inject 1–3 hours before bed for that reason; the subQ timing itself has never been tested.

5
Rotate around the abdomen

Inject subcutaneously, moving the spot each night. Keep 2 inches from the navel and away from scars or bruises.

6
Back in the fridge

Store the mixed vial at 2–8 °C and plan to finish it in about 28 days, the window bacteriostatic water is usually given. No DSIP-specific stability data has been published.

Feeling Something? Check Here

🟢 Normal: keep going
  • Nothing you can feel on the first night or two.
  • Feeling slightly more awake for an hour after the shot (seen in the trials).
  • A brief sting or small pink bump at the injection site.
  • Vivid dreams; often reported, never measured.
🟡 Stop and get advice
  • Headache, nausea, dizziness or sweating after doses (reported in the withdrawal trials).
  • Morning grogginess that lasts into the day.
  • Sleep getting worse instead of better by week 2.
  • Injection-site redness or lumps still there after 24 hours.
🔴 Emergency: stop immediately
  • Light-headedness or fainting: 2 of 107 people in one trial had a drop in blood pressure.
  • Swelling of the face, lips or throat, wheeze, or spreading hives.
  • Fever with spreading heat, redness or pus at an injection site.
Do not use DSIP if any of these apply
  • Pregnancy, trying to conceive, or breastfeeding: no data of any kind.
  • Under 18.
  • Untreated sleep apnea: loud snoring, gasping or daytime sleepiness need a sleep study, not a sleep peptide.
  • Low blood pressure, or blood-pressure medication (the trial cases of hypotension).
  • ACE-inhibitor use such as captopril: the enzyme those drugs block breaks DSIP down, so levels could change (a theory, not tested).
  • Alcohol, benzodiazepines, Z-drugs or other sedatives the same night.
  • As a replacement for prescribed insomnia, anxiety or withdrawal treatment.

DSIP Dosage FAQ

How much DSIP should I take for sleep?+

Most people use 100–300 mcg injected under the skin in the evening, with 250 mcg the most common. No study ever tested those numbers. Every human trial gave DSIP by IV at 25–30 nmol/kg, which works out to about 1.5–1.8 mg for a 70 kg adult, and the FDA said in 2026 that it found no data at all for subcutaneous use. Start at 100 mcg, keep a sleep log, and judge it over two weeks.

How long before bed should I take DSIP?+

In the trials, DSIP was given by IV immediately before bed or about an hour before, and its sleep effect started in the second hour after the injection, after a slightly alert first hour. That is why many people inject 1–3 hours before bed. Morning injections also improved the following night in one series, so the exact timing may matter less than consistency.

What is the half-life of DSIP?+

Very short. Human serum breaks it down in 5–10 minutes in the test tube, and the FDA briefing gives a plasma half-life of about 8 minutes. In dogs, rats and monkeys it is 2–4 minutes. The sleep effects in the trials outlasted the drug, including better sleep the next night, which is why it is dosed once a day and not more often.

Does DSIP actually work?+

The evidence is mixed. Small 1980s trials led by one research group were positive, with total sleep up 57–97 minutes in insomniacs. The two independent placebo-controlled trials, Monti 1987 (6 people) and Bes 1992 (16 people), found little or no benefit. The FDA called the insomnia evidence "inconclusive and at best preliminary" in its 2026 review.

Can I take DSIP every night?+

Nightly dosing is what the trials did, but only for about a week (15 days at most). Over 4–7 doses the effect built up rather than faded, and no tolerance was documented. Nobody has tested months of nightly use, so most people run 2–4 weeks and then stop for at least as long.

What are the side effects of DSIP?+

The insomnia trials reported none. In 107 people treated during drug or alcohol withdrawal, 9 had minor headache, nausea, dizziness or sweating, and 3 had serious reactions, 2 of them a drop in blood pressure. The FDA adverse-event database had zero reports as of 2024. Long-term safety is unknown.

Is DSIP legal or FDA approved?+

DSIP is not an approved drug in any country. In July 2026, an FDA advisory committee voted 6 to 7 against letting pharmacies compound it (it was the only one of seven peptides voted down that day), after FDA staff recommended against it. It is sold online as a lyophilized powder, mostly in 5 mg and 10 mg vials.

How do I mix a 10 mg vial of DSIP?+

Add 5 mL of bacteriostatic water for 2 mg/mL. On a U-100 syringe, 100 mcg is then 5 units, 250 mcg is 12.5 units and 300 mcg is 15 units. With only 2 mL the draws shrink to 2–6 units, which are hard to measure, and the concentration (5 mg/mL) is above the 0.5 mg/mL water solubility the FDA lists for the free-base form.

Does nasal or oral DSIP work?+

There is no sleep data for either. DSIP did not cross a layer of human gut cells in a lab test, which argues against oral use. One 2001 letter reported a brain-wave change after a nasal dose, and that is the only human non-IV signal found.
