Kisspeptin is a neuropeptide that stimulates GnRH release, driving LH and FSH secretion for natural testosterone and reproductive hormone support. Here are the most commonly used dosing protocols for subcutaneous injection.
100 mcg subcutaneously, 3x per week is the common protocol, roughly the 1 µg/kg dose that produced peak LH in healthy men. 50 mcg 2x per week to start. Not daily: with kisspeptin it is frequency, not dose, that shuts the receptor down. All of the effect data comes from intravenous delivery.
Dosage Protocols

About 0.5 – 0.7 µg/kg for most adults, inside the 0.3 – 1 µg/kg IV range that raised LH in men.
Why this dose+
Two days a week is enough to find out whether you respond at all. The catch is that 2.5 units sits halfway between marks on a U-100 syringe, so mix the vial with more water rather than eyeballing the draw.
Roughly the 1 µg/kg IV bolus that produced maximal LH in healthy men (4.1 → 12.4 IU/L).
Why this dose+
About 1.0 – 1.4 µg/kg depending on body weight: 100 mcg is ~1 µg/kg at 100 kg and ~1.4 µg/kg at 70 kg. Treat it as a ceiling, not a midpoint. The next dose step up in that trial produced no significant LH rise, so lighter adults have no headroom above this.
The only long-run kisspeptin schedule with human support, and where you drop to if a 3x/week block fades.
Why this dose+
Twice-weekly kisspeptin-54 kept reproductive hormones elevated across 8 weeks in women with hypothalamic amenorrhoea, while twice-daily dosing caused tachyphylaxis. Fewer doses per week is the corrective move when a response fades, not a bigger dose.
- LH tripling, testosterone climbing 45%, pulse frequency normalising: all IV bolus or infusion.
- The one published subcutaneous trial found no gonadotrophin response at doses up to 32 nmol/kg.
- The ~4-minute half-life is the likely reason; kisspeptin-54 (27.6 min) is the form used subcutaneously.

Alcohol swab the kisspeptin vial and the bacteriostatic water vial, and let both air-dry before you pierce them.
Doses here are 50 – 100 mcg, which is tiny. A 5 mg vial in 2.5 mL gives 2,000 mcg/mL, so 100 mcg is a readable 5 units. Reconstitute thin and the maths stops being guesswork.
Run the water down the inside wall of the vial, then roll it gently until the cake dissolves and the solution is clear. Shaking damages the peptide.
Use a fresh U-100 insulin syringe. Pull to the unit mark from the calculator and flick out any air bubbles.
Pinch a fold of abdominal fat and rotate sites between doses, staying clear of the navel, scar tissue and bruises. LH peaks 30 – 45 minutes later, so morning keeps the pulse where your own axis expects it.
Keep lyophilised vials in the freezer and away from light. Once mixed, refrigerate and use within about 4 weeks. Never freeze a reconstituted vial. Needles go in a sharps container.
The four-week refrigerated window is generic peptide handling, not a published kisspeptin-10 stability figure. If a vial would last you months at 2 – 3 doses a week, mix a smaller volume more often.
What to Expect
Almost everything kisspeptin does happens on a lab report rather than in how you feel. The timings below come from intravenous human studies; the honest way to know your own protocol is working is a blood draw, not a sensation.
A single IV dose in healthy men lifted LH from 4.1 to 12.4 IU/L, peaking by 45 minutes. You will not notice this happening; a blood draw in this window is the only real test.
In men with type 2 diabetes and mild hypogonadism, an 11-hour infusion took testosterone from 8.5 to 11.4 nmol/L and raised LH pulse frequency from 0.6 to 0.9 pulses/hour. In healthy men, a 22.5-hour infusion moved testosterone from 16.6 to 24.0 nmol/L. Both required continuous IV delivery; a single subcutaneous shot has no comparable data behind it.
Libido and more frequent morning erections are the most commonly reported first signs. The strongest human evidence uses a different molecule and route: kisspeptin-54 infused IV at 1 nmol/kg/h for 75 minutes in men with hypoactive sexual desire disorder modulated sexual brain processing and increased penile tumescence to visual sexual stimuli by up to 56% versus placebo.
Twice-weekly kisspeptin dosing sustained reproductive hormone release across 8 weeks in women with hypothalamic amenorrhoea; twice-daily dosing did the opposite. If your response is fading here, the corrective move is fewer doses per week, a break, or both.
Kisspeptin-10 stores nothing and leaves nothing behind. LH and testosterone return to whatever your own axis was producing before, typically within days rather than weeks. There is no reported crash or rebound suppression from intermittent use at these doses, but equally, nothing carries over.
Kisspeptin-10 acts on your own pituitary, so its effect is capped by the axis underneath it. Sleep, enough calories and body fat to run a reproductive axis, and a workup for any real cause of low LH all raise that ceiling.

- New acne, oily skin, irritability or mood swings: a downstream androgen shift.
- Testicular ache or swelling.
- Cycle irregularity, breakthrough bleeding or a missed period in women.
- Nipple tenderness or breast tissue changes in men.
- Allergic reaction: hives, swelling of lips or tongue, wheeze, throat tightness.
- Chest pain, fainting, or a racing heart after a dose.
- Severe pelvic pain, abdominal bloating and swelling, rapid weight gain or reduced urine output: ovarian hyperstimulation.
- OHSS is a real risk alongside any fertility stimulation, since kisspeptin-54 is used clinically as an ovulation trigger.
- Sudden vision changes or a severe unrelenting headache: a pituitary red flag.
- Fever, spreading redness, warmth or pus at an injection site.
- Any hormone-sensitive cancer: prostate, breast, testicular, or any tumour driven by LH, testosterone or oestrogen.
- Pregnancy or breastfeeding: circulating kisspeptin rises thousands-fold in pregnancy, and dosing on top of that is untested.
- A GnRH agonist or antagonist (leuprolide, goserelin, degarelix), or androgen-deprivation therapy, which kisspeptin works directly against.
- An ongoing IVF, ovulation-induction or fertility-stimulation cycle, unless a fertility specialist is directing it.
- Kisspeptin can trigger a premature LH surge and precipitate OHSS.
- Known hypersensitivity to the peptide, or to any excipient or preservative in the vial.
- An age under 18, or delayed or incomplete puberty, outside specialist endocrine care.
- Undiagnosed low testosterone, amenorrhoea or infertility that has never been worked up.
- Prolactinoma, pituitary adenoma, haemochromatosis and thyroid disease all present that way and need finding first.
- A known pituitary tumour, or prior pituitary surgery or radiotherapy.
Kisspeptin-10 is not an approved drug anywhere. FDA's Pharmacy Compounding Advisory Committee voted in October 2024 against adding it to the 503A bulks list, so no US pharmacy can legally compound it.
FAQ
Is kisspeptin-10 the same thing as kisspeptin-54?+
No, and the difference matters more than almost anywhere else in peptides. Kisspeptin-10 is the active C-terminal 10 amino acids of kisspeptin-54 (metastin), so the receptor pharmacology is the same, but the plasma half-life is roughly 4 minutes versus 27.6 minutes. Matched molar IV infusions produce comparable gonadotrophin responses, even though kisspeptin-54 reaches up to 37-fold higher circulating levels. Critically, nearly every headline human result attributed to "kisspeptin" used kisspeptin-54: the sexual brain-processing and HSDD trials, the IVF oocyte-maturation trigger work, the 8-week hypothalamic amenorrhoea study, and all the successful subcutaneous dosing. Kisspeptin-10 is the cheaper, shorter-acting molecule, and it is not a drop-in substitute.
Will it actually raise my testosterone?+
In men whose problem is upstream (low LH driving low testosterone), yes, modestly, and only while dosing. An 11-hour IV infusion in men with type 2 diabetes and mild hypogonadism took testosterone from 8.5 to 11.4 nmol/L, and a 22.5-hour infusion in healthy men moved it from 16.6 to 24.0 nmol/L. Both needed continuous infusion. If your testes cannot respond (primary hypogonadism, prior injury, Klinefelter’s) or you are already on exogenous testosterone, kisspeptin has nothing to push on and will do essentially nothing. Get LH, FSH, total and free testosterone, prolactin and SHBG measured before you decide this is your problem.
Can I use it instead of hCG or enclomiphene to restart my axis after a cycle, or on TRT?+
There is no human trial of kisspeptin-10 for HPG-axis restart, so anyone offering you a validated restart protocol is ahead of the data. Mechanistically it sits above GnRH, so it needs an intact hypothalamus, pituitary and testes to work; hCG bypasses that whole chain and acts directly on the Leydig cell, which is why it is the tool with actual evidence behind it. On TRT it is the least promising option of all: exogenous testosterone suppresses at the level of the hypothalamus and pituitary, which is precisely the machinery kisspeptin would have to work through. Talk to a physician about hCG, enclomiphene or an SERM protocol instead.
Why not just take it every day, or twice a day?+
Because the KISS1R receptor does not tolerate continuous stimulation. Twice-daily kisspeptin-54 in women with hypothalamic amenorrhoea caused LH responsiveness to fade progressively and FSH responsiveness to be nearly abolished by day two, whereas twice-weekly dosing produced only partial desensitisation and sustained hormone release over eight weeks. The extreme version proves the point: TAK-448, a kisspeptin analogue formulated for sustained exposure, drove testosterone into the castration range and was developed as a prostate cancer drug. Pulsatile, intermittent dosing is not a conservative preference here; it is the mechanism.
How much bacteriostatic water should I use, and does it really matter?+
More than you would use for most peptides, and yes it matters a lot. Doses here are 50 – 100 mcg, which is tiny. A 5 mg vial in 2.5 mL of bacteriostatic water gives 2,000 mcg/mL, so 100 mcg is 5 units on a U-100 insulin syringe and 50 mcg is 2.5 units. If you instead reconstitute a 10 mg vial in 1 mL, 100 mcg becomes 1 unit, where a half-unit misread is a 50% dosing error. Use the most water your vial will physically hold, keep the mixed vial in the fridge, and use it within about four weeks. The extra diluent costs nothing and the accuracy is free.
Does it work for libido?+
The best human evidence is genuinely encouraging but does not describe what you would be doing. In a placebo-controlled crossover trial in men with hypoactive sexual desire disorder, kisspeptin-54 infused intravenously at 1 nmol/kg/h for 75 minutes modulated activity in sexual-processing brain regions and increased penile tumescence to visual sexual stimuli by up to 56% versus placebo. That is a real result with a real control group. But it is a different molecule, a different route, and an acute effect measured inside the study session; it is not evidence that weekly subcutaneous kisspeptin-10 improves desire over time, and no trial has tested that.
Do women respond differently?+
Dramatically, and the difference is not subtle. Healthy women in the follicular phase showed no gonadotrophin response to kisspeptin-10 at all: not to IV bolus up to 10 nmol/kg, not to subcutaneous bolus up to 32 nmol/kg, not to IV infusion. In the preovulatory phase, the same IV bolus raised both LH and FSH. Post-menopausal women are more responsive than cycling women, and women on hormonal contraception respond minimally. The response tracks baseline oestradiol closely: in one subcutaneous kisspeptin-54 study, a 100 pmol/L higher baseline oestradiol predicted about 1.0 IU/L more LH response. In practice, where a woman is in her cycle changes the answer more than the dose does.
Is it legal or FDA-approved?+
No, on both counts. Kisspeptin-10 is not an approved drug in any jurisdiction. FDA’s Pharmacy Compounding Advisory Committee reviewed it on 29 October 2024 and voted against adding it to the 503A bulks list (alongside ipamorelin, ibutamoren and L-theanine), citing insufficient human safety data and inadequate efficacy evidence, so US compounding pharmacies cannot legally use it. It was also not among the twelve peptides (BPC-157, TB-500, Epitalon and others) that FDA removed from Category 2 in April 2026. Everything on the market is sold "for research use only", which means nobody is verifying identity, purity or sterility on your behalf; third-party testing is on you.
